Recerca de selenoproteïnes en el genoma de protistes: GPx i FrnE


La utilització excepcional del codó UGA per codificar selenocisteïnes (Sec) fa molt difícil la identificació de les selenoproteïnes en els genomes, donat que pràcticament tots els programes bioinformàtics assumeixen que aquest codó codifica per una senyal de terminació de la traducció. Només quan el codó UGA precedeix un element SECIS a l'extrem 3'UTR i l'organisme disposa de la maquinària de traducció i inserció específica i necessària, codificarà per una selenocisteïna.


És per això que la supervisió humana i l'ús d'altres mètodes, com ara la identificació d'aquests elements SECIS i l'anàlisi de la presència o no de la maquinària requerida, són necessaris per la caracterització de selenoproteïnes en els diferents genomes.


L'objectiu del nostre projecte és, doncs,la cerca i caracterització de les selenoproteïnes GPx i FrnE en 14 genomes de diferents espècies de protistes recentment seqüenciats. Per dur a terme aquesta tasca hem dut a terme estudis d'homologia amb seqüències conegudes de les selenoproteïnes assignades, de predicció de l'estructura genòmica dels homòlegs potencials i d'identificació dels possibles elements SECIS existents.


El que en podem extreure de tots aquests anàlisi, en què aprofundirem a continuació, és una àmplia distribució de les selenoproteïnes al llarg de l'evolució, fet que deixa la porta oberta a futures investigacions en aquest camp.





Research selenoproteïnes in the genome of protists: GPX and FrnE


The exceptional use of the stop-codon UGA to code for selenocysteine (Sec) makes the identification of selenoproteins in given genomes difficult, as virtually all gene-prediction programs assume this codon to be a translation termination signal. Only when the UGA codon precedes a SECIS-element in it's 3'UTR, and the organism disposes of the specific selenocysteine synthesis and insertion machinery, it codes for selenocysteine.


For that reason, human supervision is beneficial and the usage of further methods, like SECIS prediction and testing for the existence of the required machinery, are needed for the correct annotation of selenoproteins in different genomes.


The objective of our project is the search and characterization of the selenoproteins GPx and FrnE in 14 different newly sequenced genomes of protists. To achieve this task, we made homology studies of known sequences of the given selenoproteins, prediction of the genomic structure of potential homologs and identification of possible SECIS elements.
