RESUMEN

A continuación les presentamos un trabajo de investigación en el que pretendemos caracterizar selenoproteínas en un grupo de microorganismos en el que todavía no se conoce su existencia, los protistas. Los protistas son un grupo taxonómico que presenta una gran diversidad con respecto a estas proteínas, cuya distribución no está estudiada aún en detalle.

Las selenoproteínas incorporan un aminoácido de selenocisteína. Contienen, por lo tanto, una secuencia de inserción de selenocisteína llamada SECIS en su extremo 3' UTR.

La dificultad de la identificación de selenoproteínas recae en el hecho de que el aminoácido de selenocisteína es codificado por el codón UGA, que normalmente es interpretado por la mayoría de sistemas de secuenciación como un stop de la traducción. Por esta razón, la anotación de este tipo de proteínas no es muy exhaustiva, y en muchos organismos aún no se han analizado.

Pretendemos, mediante el uso de herramientas bioinformáticas, lograr encontrar la presencia de selenoproteínas en el genoma de estos microorganismos y de sus homólogos en cisteína, consiguiendo así mejorar la anotación de su secuencia aminoacídica. En concreto, nos centraremos en el estudio de la familia de las GPX en el genoma de los protistas secuenciados durante el 2010.




ABSTRACT


In the current study, we present a research study in which we try to characterise selenoproteins in a group of organisms, the protists.

Protist are a taxonomical group which presents a high diversity regarding this kind of proteins. In addition, the distribution of selenoproteins in this organisms have not been carefully studied . Selenoproteins have a selenocisteine aminoacid in their sequence. Consequently, they have an insertion sequence at the 3' position called SECIS.

What makes the study more difficult is the fact that UGA codon, the one that codifies for the selenoprotein, is usually understood as an stop codon by the majority of sequencing systems. For this reason, there is not much information about this proteins and not many research groups have looked for them in certain organisms such as the protists.

The objective of our study is to find, thanks to the bioinformatical tools , some members of the GPX selenoproteins (and their cisteine homologs) in the 11 genomes which have been sequenced in the current year 2010.
